چکیده

Polycyclic aromatic hydrocarbons (PAHs), products from the incomplete combustion of crude oil, are pollutants present in nature. Ring hydroxylating dioxygenase enzymes able to catalyze polycyclic biodegradation process with a high degree stereo-, regio-, and enantiospecificity. In this work, we first approximation binding modes 9 PAHs aromaticity catalytic sites biphenyl or naphthalene dioxygenases four microorganisms usually used bio-remediation processes: Sphingobium yanoikuyae, Rhodococcus jostii RHA1, Pseudomonas sp. C18, Paraburkholderia xenovorans. Molecular modeling studies two yanoikuyae xenovorans showed good affinity for 2–4 benzene rings (fluoranthene, pyrene, chrysene), both had similar amount substrate binding. docking using C18 that enzyme is accommodate (benzo(a)pyrene, indeno(1,2,3-cd)pyrene), scores. This study provides important insight into utility degradation HAPs aromaticity.

SEPARATION OF POLYCYCLIC AROMATIC HYDROCARBONS WITH MIXED MICELLAR HIGH PERFORMANCE LIQUID CHROMATOGRAPHY

Separation of Polycyclic Aromatic Hydrocarbons was performed by RP-HPLC using mixed micellar mobile phase. Effect of temperature, types and amounts of coorganic solvent in mobile phase on efficiency, asymmetry factor and selectivity factor showed that the appropriate condition for separation of these compounds was 3% isopropanol (V/V) as co-organic solvent in mobile phase at 40°C. Analysis of...

Identification and functional analysis of two aromatic-ring-hydroxylating dioxygenases from a sphingomonas strain that degrades various polycyclic aromatic hydrocarbons.

In this study, the enzymes involved in polycyclic aromatic hydrocarbon (PAH) degradation in the chrysene-degrading organism Sphingomonas sp. strain CHY-1 were investigated. [14C]chrysene mineralization experiments showed that PAH-grown bacteria produced high levels of chrysene-catabolic activity. One PAH-induced protein displayed similarity with a ring-hydroxylating dioxygenase beta subunit, an...

Biodegradation of Polycyclic Aromatic Hydrocarbons (PAHs) in Petroleum Contaminated Soils

Polycyclic aromatic hydrocarbons are a class of potentially hazardous chemicals of environmental and health concern. PAHs are one of the most prevalent groups of contaminants found in soil. Biodegradation of complex hydrocarbon usually requires the cooperation of more than single specie. In this research biotreatment of PAH (phenanthrene) was studied in a solid-phase reactor using indigenou...

Transport of Polycyclic Aromatic Hydrocarbons in a Calcareous Wetland Soil

Knowledge of transport and degradation of polycyclic aromatic hydrocarbons (PAHs) is important in assessing PAH contamination of soils and water resources. The transport of naphthalene, anthracene, pyrene and phenanthrene was determined in a contaminated calcareous soil obtained from the Shadegan wetland (Khozestan, Iran) considering a column study in laboratory conditions. The PAHs were added ...
